These changes include blebbing, cell shrinkage, nuclear fragmentation, chromatin condensation, chromosomal DNA fragmentation, and global mRNA decay. A failure of cells to undergo apoptosis is a common feature of many cancers thus investigation of apoptosis inducing mechanisms is of particular importance for cancer research and requires suitable detection methods.

A hallmark of late apoptosis is extensive genomic DNA fragmentation that generates a multitude of DNA double-strand breaks with accessible 3'-hydroxyl (3'-OH) groups. Terminal deoxynucleotidyl transferase dUTP Nick End Labeling (TUNEL) assay is a method for detecting DNA fragmentation by labeling the 3’-OH termini in the double-strand DNA breaks generated during apoptosis. The TUNEL assay relies on the presence of nicks in the DNA which can be identified by TdT, an enzyme that catalyzes the addition of dUTPs that are labeled with fluorescein.
